چکیده

Proteomics is the study of the collection of proteins (the proteome) expressed in a given system. The field of proteomics can be divided into three subtypes: • profiling – identification and quantification of the proteins in a sample and drawing comparisons between samples, • functional – the study of the behavior of specific proteins, and • structural – the study of protein folding and the complexes it forms. Microfluidics is characterized by devices that contain channels or features with dimensions measured in microns, and are capable of transporting and analyzing sample volumes in the nanoliter range.

Hydrogel Discs on Digital Microfluidic Devices for Proteomic Applications

We introduce the use of immobilized enzymes in hydrogels for proteomic sample processing in digital microfluidic systems. In this technique, pre-formed cylindrical agarose discs bearing immobilized enzymes were integrated into digital microfluidic devices bearing arrays of electrodes. In some experiments, fluorogenic enzyme substrates were delivered to gel discs which served as integrated micro...

Electrochemical conversions in a microfluidic chip for xenobiotic metabolism and proteomics

Albert van den Berg is a full professor on Miniaturized Systems for (Bio)Chemical Analysis at at the University of Twente. In this contribution he describes how microfluidic techniques can be used to mimic xenobiotic metabolism in vitro. Similar devices can also be used to electrochemically cleave proteins for mass spectrometric detection and database searching.

Microfluidic single-cell analysis of intracellular compounds.

Biological analyses traditionally probe cell ensembles in the range of 103-106 cells, thereby completely averaging over relevant individual cell responses, such as differences in cell proliferation, responses to external stimuli or disease onset. In past years, this fact has been realized and increasing interest has evolved for single-cell analytical methods, which could give exciting new insig...

Multidisciplinary Role of Microfluidics for Biomedical and Diagnostic Applications: Biomedical Microfluidic Devices

Life scientists are closely working with engineers to solve biological and biomedical problems through the application of engineering tools. For engineers involved in this collaborative work, new knowledge is created in their own disciplines. For example, the science and technology at the interface of biomedical sciences and microfluidics has played a significant role in ushering in recent adva...

Microfluidic Protein Separation by Free Flow Isoelectric Focusing

Disposable, inexpensive microfluidic devices have the potential to become a robust tool for proteomic research for separating and identifying proteins and protein complexes. In this work, a preparative scale free flow IEF isoelectric focusing (FFIEF) device was designed, investigated, and optimized. A constant electric field of up to 350V/cm was generated across the separation channel upon opti...
